[edit] MECHANISM OF ACTION

Griseofulvin binds microtubules (depolymerizes) and prevents spindle formation during mitosis. It is fungistatic.

Griseofulvin has a particular affinity to Keratin so it accumulates in keratinized layer of epithelium, hair and nails. Since insoluble, it is administered as suspension, and absorption can be inceased by fatty meals.

Oral therapy for topical fungal infections. it is only active against dermatophytes

Contraindicated in pregnancy because of suspected teratogenicity

[edit] SIDE EFFECTS

  • Photosensitivity
  • CNS: headache (up to 15%), confusion, insomnia, memory impairment, fatigue, blurred vision and sometimes parathesias or psychosis
  • GI: Upset stomach, nausea, vomiting and diarrhea
  • Hematological effects: leukopenia, neutropenia, monocytosis, basophilia. Blood test should be carried out weekly for 1st month of treatment
